The complete genetic characterization of tumours is important not only to understand tumour biology, but also to the development of new drugs and to the selection of patients that may benefit of a given targeted cancer therapy. The promise of using proteins encoded by mutant cancer genes as molecular targets for the development of novel therapies drives endeavours to identify novel mutated cancer genes as well as to create catalogues of somatic mutations in cancer. Our current projects in the laboratory focus in this direction.

Thus, the main goals of the group include:

To identify novel genes altered, at a genetic level, in cancer.

Our group is interested in the identification of novel cancer genes following the screening for genes that participate in relevant and novel cancer pathways and using high through put technologies such as CGH arrays and next generation sequencing approaches.

To understand how these abnormalities contribute to cancer development.

In addition to identify novel cancer genes, we are also interested in functionally analyze them by inactivating their expression using RNA interference in either tumor or normal cells and then studying morphological and biochemical changes. Alternatively, transfecting wild type copies into human cancer cells and testing for their ability to suppress malignancy in vitro and in vivo as well as characterizing the biological pathway to which they belong.

To identify molecular markers that determine sensitivity to novel cancer drugs.

Identifying novel genes mutated in cancer and understanding their functions and the pathways disrupted provides a foundation for a translational research. These will be essential for the validation of novel cancer genes and their biological pathways and for study of their relevance in differential diagnosis and patient prognosis. Moreover, through collaborations with Oncology Departments that participate in clinical trials, we envision our lab to work in the test of molecular markers that may confer resistance to novel therapeutic agents.

Montse Sanchez-Cespedes nació en Badalona (Barcelona) en 1968. Licenciada en Biología en la especialidad de Genética y Biología Molecular en la Universidad de Barcelona, realizó su trabajo predoctoral en el Laboratorio de Biología Molecular del Cáncer del Hospital ”Germans Trias i Pujol” de Badalona. De 1997 a 2001, fue Investigadora Posdoctoral en la Johns Hopkins University School of Medicine (Baltimore-USA). Centró su trabajo en la identificación de nuevas alteraciones genéticas y moleculares del cáncer, en concreto, en la búsqueda de   alteraciones cromosómicas recurrentes en tumores de pulmón, así como en la identificación de nuevos genes supresores de tumores en esas regiones. Entre los logros más importantes alcanzados durante ese período destaca el descubrimiento de la inactivación frecuente del gen supresor tumoral LKB1 en adenocarcinomas de pulmón.

De 2001 a 2008 fue jefe del Grupo de Cáncer de Pulmón del Centro Nacional de Investigaciones Oncológicas (CNIO), desde donde hizo contribuciones importantes en el área de la genética del cáncer de pulmón. Además de seguir las investigaciones acerca del papel de LKB1 en este tipo cáncer, la Dra. Sanchez-Cespedes llevó a cabo estudios de los perfiles de expresión y de alteraciones en el número de copias del DNA, así como la búsqueda de nuevos genes involucrados en cáncer de pulmón. En el CNIO, su grupo describió la presencia de mutaciones comunes en otro gen supresor tumoral denominado BRG1, un componente del complejo de remodelador de cromatina SWI/SNF. Desde Octubre del 2008, la Dra. Sanchez-Cespedes dirige el Grupo de Genes y Cáncer en el Programa de Epigenética y Biología Molecular del Instituto de Investigación Biomédica de Bellvitge (IDIBELL) en Barcelona.

Entre otros reconocimientos a la Dra. Sánchez-Céspedes, se encuentran un contrato Ramón y Cajal obtenido en el 2001 y el reconocimiento por la Generalitat de Catalunya de Grupo Consolidado otorgado a su grupo actual en el PEBC. Su lista de más de 60 publicaciones y artículos originales en revistas internacionales acreditadas de prestigio sirven de testimonio de la experiencia que ha ido acumulando en el campo de la investigación del cáncer.
